Damnacanthus major is a shrub and has a low biomass, making it difficult to use as a cosmetic ingredient. Therefore, we secured biomass through callus culture to solve the problem. Previously in our study, calli of Damnacanthus major treated with elicitor had antioxidant activity. Methyl jasmonate (MeJA) is one of the abiotic elicitors that increases plant useful substances in culture. Damnacanthus major callus was harvested by culturing for 2 weeks in a 250ml flask or 5L bioreactor and then treating MeJA for 1 week. Fresh weight was highest in the group without MeJA at 52.3 g and 864.0 g in both culture scales (250 ml flask and 5 L bioreactor), respectively. Dry weight was highest in the untreated group (2.0g, 32.7g). Fresh and dry weight, growth index tended to decrease depending on MeJA concentration. Dry matter rate also showed a similar trend in both culture scales, and the 200 μM MeJA treated group was about 1.4 times higher than the untreated group. Digiperuginol-11-O-β-primeveroside, a new molecules not found in wild plant leaves, was isolated from callus extract. When treated with MeJA 100μM, the amount of new material was approximately 46 times higher than when treated without MeJA. ABTS scavenging activity increased depending on the concentration of single molecules. Based on these results, it is assumed that Damnacanthus major cultured elicitor will be capable of pilot-scale culture and can be used as an cosmetic ingredient.
